[Indications] It is a kinase inhibitor suitable for the treatment of Adult patients with chronic phase accelerated phase or blastic phase chronic myelogenous leukemia who are resistant to or intolerant to previous tyrosine kinase inhibitor therapy or D chromosome positive who are resistant to or intolerant to previous tyrosine kinase inhibitor therapy Acute lymphoblastic leukemia (+). This indication is based on response rate. There are no useful trials demonstrating improvement in disease-related symptoms or increased survival.
[Usage and Dosage] Take orally once a day with or without food.
[Precautions]
Congestive heart failure: Monitor patients for signs and symptoms of congestive heart failure and treat when clinically indicated.
Hypertension: Monitor high blood pressure and treat when clinical indications indicate.
Pancreatitis: Monitor serum esterase monthly for interruption or discontinuation.
Bleeding: Interrupt for severe bleeding
Fluid Retention: Monitor patient for discontinuation of fluid retention.
Arrhythmia: Monitor for symptoms of arrhythmia.
Myelosuppression: Thrombocytopenia, neutropenia, and anemia may require dose interruption or reduction. Monitor complete cell counts biweekly for 1 month and then monthly and as clinically indicated. On/or thrombocytopenia/interruption.
Tumor lysis syndrome: Ensure hydration and correction of high uric acid levels before initiating treatment
Impaired wound healing and gastrointestinal perforation: Briefly interrupt treatment in patients undergoing major surgery.
Embryo-fetal toxicity: May cause fetal harm. Advise women of the potential risk to the fetus.
[Adverse Reactions] The most common non-hematological adverse reactions (%) are high blood pressure, rash, abdominal pain, fatigue, headache, dry skin, constipation, joint pain, nausea, and fever. Hematologic adverse reactions include thrombocytopenia, anemia, neutropenia, lymphopenia, and leukopenia.
